  • Consult Authoritative Databases: The ATCC (American Type Culture Collection) website provides detailed culturing information for most cell lines, including recommended media, conditions, and related literature.

II. Storage and Shelf Life

2. Can liquid medium be stored at -20°C?

  • No. Storage at -20°C can cause salts in the medium to precipitate. Upon thawing, these salts may not fully redissolve, leading to a decrease in osmolarity. This can cause cells to rupture and die.

3. How long can newly prepared liquid medium be stored?

  • It is recommended to store newly prepared medium at 4°C protected from light and use it within one week. The fresher the medium, the better.

4. Can medium supplemented with serum and antibiotics be stored long-term?

  • No. Once serum and antibiotics are added to fresh medium, it should be used within two to three weeks. Some antibiotics and essential components in serum begin to degrade after thawing.

5. Why does medium stored in the refrigerator turn dark red (alkaline)?

  • When stored at 4°C, CO₂ gradually escapes from the medium, causing the pH to increase (become more alkaline). The pH indicator phenol red turns a darker red-purple under alkaline conditions.
  • Solution: The pH can be adjusted by bubbling filter-sterilized CO₂ through the medium before use.

  • NaHCO₃ (Sodium Bicarbonate): Forms a buffering system with CO₂ to maintain stable pH in the incubator.

  • HEPES: A potent chemical buffer that provides additional pH stabilization, especially outside the CO₂ incubator environment. Common final concentration is 25 mM.
  • Phenol Red: Acts as a pH indicator (Red = Neutral ~7.4, Yellow = Acidic, Purple = Alkaline).
  • L-Glutamine: An essential amino acid for cells, involved in energy metabolism and protein synthesis. It is unstable in solution and must be stored frozen at -20°C. It is typically added to medium just before use. Common final concentration is 2 mM. Medium supplemented with glutamine should be re-supplemented if stored at 4°C for longer than 2 weeks.

  • Note: The pH of liquid often increases by ~0.2 units after filtration through a 0.1µm or 0.2µm filter.

11. Why does our RPMI 1640 medium appear yellow? Is the pH wrong?

  • This is normal. Due to its formulation, RPMI 1640 is a low phenol red variant; it contains only one-fourth the amount of phenol red found in DMEM. The lighter, yellowish color is due to this lower concentration, not an incorrect low pH.

RPMI-1640 Medium

RPMI stands for Roswell Park Memorial Institute, and RPMI is a class of cell culture media developed by the Institute, while RPMI 1640 is a more specific type of cell culture media.

Originally developed for the suspension or monolayer culture of human leukemia cells, RPMI-1640 medium has since been found suitable for a wide range of mammalian cells, including HeLa, Jurkat, MCF-7, PC-12, PBMC, astrocytes, and cancer cells. It is particularly suitable for the culture of suspension cells. RPMI-1640 medium is one of the most widely used media.

RPMI 1640 medium contains the reducing agent glutathione, high concentrations of vitamins, biotin, vitamin B12, and PABA, and high concentrations of vitamins inositol and choline. RPMI-1640 medium contains no proteins, lipids, or growth factors; therefore, RPMI-1640 requires supplementation, usually with 10% fetal bovine serum (FBS).

DMEM (Dulbecco’s Modified Eagle Medium)

DMEM is based on Eagle’s minimum essential medium MEM, which is divided into two types: high glucose (containing 4500 mg/L glucose) and low glucose (containing 1000 mg/L glucose).

The low glucose type Eagle “s Minimum Essential Medium is suitable for dependent adherent cell culture, especially for tumor cell culture with a fast growth rate and poor adhesion.

High glucose DMEM is more suitable for high-density suspension cell culture and clonal culture with poor adhesion but does not want to be detached from the original growth point, it also can be used for hybridoma myeloma cells and DNA transfected transformed cell culture. Others, such as M199, IMDM, and L15 medium, are also used to culture certain cells. The characteristics and applications of these cell culture media are as follows.

1. BME cell culture medium

Basic Eagle medium was designed by Eagle in 1955, containing BSS + 12 kinds of amino acids + glutamine + 8 kinds of vitamins. It is simple, easy to add, and suitable for various passaged cell lines and special research use, based on which improved varieties of cell culture medium are MEM, DMEM, IMDM, etc.

2. MEM cell culture medium

This is improved on Eagle’s basic medium (BME) in 1959, lysine and biotin were removed, and amino acid concentration was increased. However, because of its limited nutrient content, it is not necessarily the best medium for production-specific cell culture.

3. DMEM cell culture medium

DMEM is an Eagle medium modified by Dulbecco, initially designed for mouse fibroblasts. DMEM has twice the amino acid concentration of MEM, four times the vitamin concentration of MEM, and uses double the HCO3- and CO2 concentration for better buffering. The original formulation contained 1000 mg/L of glucose, which was later adjusted to 4500 mg/L for the growth of certain cells, often referred to as low and high glucose.

Low sugar is suitable for dependent adherent cell culture, especially for tumor cell culture with a fast growth rate and poor adhesion. High sugar is more suitable for high-density suspension cell culture, for clonal culture with poor attachment but does not want it to be detached from the original growth point, and for the culture of myeloma cells and transformed cells transfected with DNA in hybridoma.

4. IMDM cell culture medium

Guilber and Iscove modified Dulbecco’ Medium to Iscove’s Medium for culturing erythrocytes and macrophage precursors. IMDM cell culture medium contains selenium, additional amino acids and vitamins, sodium pyruvate, and HEPES and replaces iron nitrate with potassium nitrate.IMDM also promotes the growth of mouse B lymphocytes, LPS-stimulated B cells, bone marrow hematopoietic cells, T cells, and lymphoma cells.IMDM is a very nutrient-rich culture medium and can be used for the rapid proliferation of high-density cells.

5. RPMI-1640 cell culture medium

Developed by Moore et al. at Roswell Park Memorial Institute in 1967, it is designed for lymphocyte culture and contains BSS + 21 kinds of amino acids + 11 kinds of vitamins. It is now also used for suspension cell culture, such as mammalian, special hematopoietic cells, normal or malignantly proliferating leukocytes, and hybridoma cells. Other adult lymphocytes like K-562, HL-60, Jurkat, Daudi, IM-9, T-cell lymphoma cells, and HCT-15 epithelial cells can be used for reference.

6. HamF10 cell culture medium

1963 Ham design, containing trace elements, can be used when the serum content is low, suitable for cloned culture. f10 is suitable for hamster, and human diploid cells, especially suitable for amniotic fluid cell culture.

7. DMEM/F12 cell culture medium

Ham’s F12 is designed for cloning CHO cells at low serum concentration and is now widely used in the analysis of clone formation rate and primary culture. F12 can also be used with DMEM and other volumes to obtain a high concentration and composition of diverse products. This medium has been applied to many primary culture, and more difficult to raise the culture of cell lines. It is also often used as a base medium for serum-free cultures because of its rich nutrient content and the ability to use less serum.

8. M199 cell culture medium

M-199 medium is a cell culture medium with defined chemical composition designed by Morgan et al. in 1950. It contains 53 components besides BSS and is a comprehensive medium used primarily for chick embryo fibroblast culture. m-199 medium must be supplemented with the serum to support long-term culture. It can be used to culture cells of many species of origin and can culture transfected cells, and is now widely used for virology and vaccine production.

9. McCoy5A medium

McCoy5A medium is a cell culture medium designed by MeCoy in 1959 for sarcoma cells and contains BSS and 40 other components. It can support the growth of various primary grafts (such as bone marrow, skin, lung, and spleen) and is mainly used for tissue biopsy culture, some lymphocyte culture, and growth support of some difficult to culture cells, in addition to general primary cell culture—for example, Jensen rat sarcoma fibroblasts, human lymphocytes, HT-29, BHL-100, and other epithelial cells.

10. L15 Cell Culture Medium

The L-15 culture medium is suitable for the culture of rapidly proliferating tumor cells and is used to culture tumor cell lines in the absence of CO2. This culture medium uses a phosphate buffer system with a further improved amino acid composition and glucose replaced by galactose.

How to choose a cell culture medium?

  1. You can check the reference literature or consult it when you buy cell lines. You can also search on the websites of some biological companies, such as the Invitrogen website, which has a Cell Line Database tool. Select the cell type you are interested in, and it will pop up recommended media, serum and transfection reagents, etc., sometimes with optimized transfection steps, which is very convenient.

L-glutamine (Glutamine)

L-glutamine (Glutamine) is an essential nutrient element in cell culture, but it is unstable in solution and will degrade spontaneously. The amount of L-glutamine can be arbitrarily adjusted according to the research needs of the medium without L-glutamine. The addition of fresh L-glutamine or its substitutes at the time of use is more beneficial to cell growth.

L-alanyl-L-glutamine (Ala-Glu), alanyl-glutamine, and alanyl-glutamyl dipeptide, is an advanced cell culture additive that can directly replace L-glutamine in the cell culture medium. l-Glutamine is an essential nutrient in cell culture, but it is unstable in solution and will, however, it is unstable in solution and spontaneously degrades to produce ammonia and pyroglutamic acid, of which ammonia is harmful to cells, whereas L-alanyl-L-glutamine is very stable in aqueous solution and does not degrade spontaneously. It is developed by Harry Eagle on the basis of Eagle basic Medium (BEM). It is one of the most basic and applicable Medium with the widest range. It is one of the most commonly used media in animal cell culture, including HeLa, BHK-21, 293, HEP-2, HT-1080, McF-7, fibroblasts and primary rat astrocytes. L-glutamine (Glutamine) is an essential nutrient element in cell culture, but it is unstable in solution and will degrade spontaneously. The amount of L-glutamine can be arbitrarily adjusted according to the research needs of the medium without L-glutamine, and the addition of fresh L-glutamine or its substitutes at the time of use is more beneficial to the cell growth.

NEAA (Non-Essential Amino Acids)

NEAA (Non-Essential Amino Acids), which are 7 NEAAs of L-alanine, L-glutamic acid, L-asparagine, L-aspartic acid, L-proline, L-serine and glycine added to MEM medium, can reduce the side effects of cells’ own production of non-essential amino acids during cell culture and effectively promote cell proliferation and metabolism.

Glucose can be adjusted at will according to research needs, which is convenient and fast. High sugar type medium is commonly used for fast-growing cells with low adhesion, myeloma cells of hybridoma, clonal cells, transformed cells of DNA transfection, various primary virus host cells, the culture of single cells and vaccine production, such as the use of CHO cells to express EPO and production of hepatitis B vaccine. Low-sugar medium is more suitable for the culture of slower metabolizing, adherent-dependent cells.

HEPES is an excellent biological buffer, which has no toxic effect on cells. The medium with HEPES can maintain a constant pH range for a longer period of time, which can effectively prevent the cell growth from being adversely affected by large pH fluctuations of the culture medium. It can be used in CO2-free incubators.

Phenol red is used as a PH indicator in the culture medium to continuously monitor the pH of the culture medium. Phenol red makes the culture medium yellow at low PH, and purple at higher PH, and red at PH 7.2~7.4, which is most suitable for cell culture. However, phenol red also has some disadvantages. Studies have shown that phenol red can mimic the effects of steroid hormones (especially estrogen), so when using estrogen-sensitive cells (such as breast tissue), it is best to use a medium without phenol red. Phenol red can interfere with detection during flow cytometric analysis. In addition, the presence of phenol red in some serum-free medium formulations can interfere with sodium-potassium balance.

Penicillin-streptomycin Solution

Grace’s Supplementary Insect cell culture medium was originally designed to grow the cells of Antherea Eucalyptus Eucalyptus, a white star and orange moth from Australia. Grace’s supplemented insect culture medium is now widely used for the growth of Armyworm cells Sf9 and Sf21. Sf9 and Sf21 cells are routinely used in the baculovirus expression vector system (BEVS) to express recombinant proteins.
