Powdered IMDM (Iscove’s Modified Dulbecco Medium)

IMDM (Iscove’s Modified Dulbecco Medium), namely Iscove Modified Dulbecco Medium, was designed by Guilber and Iscove in 1976 to culture red blood cell precursor cells and macrophages, and is ideal for rapidly proliferating, high-density cell cultures, including Jurkat, COS-7, and Macrophages.

IMDM medium is based on DMEM medium with selenium, HEPES, sodium pyruvate, additional amino acids and vitamins, which is very nutritious, highly suitable for rapid proliferation, high-density cell culture.
IMDM medium can not only culture cells with special nutritional requirements (such as mouse B lymphocytes, LPS-stimulated B cells, bone marrow hematopoietic cells, T lymphocytes, and various hybridoma cells), but also serve as the base solution for some unique serum-free media. IMDM contains no proteins, lipids, or growth factors.
Therefore, IMDM requires supplemental nutrients, often with the addition of 10% fetal bovine serum (FBS). IMDM uses a sodium bicarbonate buffer system (3.024 g/L) and therefore requires a 5-10% CO2 environment to maintain physiological pH.

This IMDM dry powder cell culture medium is formulated with sodium bicarbonate supplementation and filtration. The detailed formulation scheme is as follows:

  1. Add 950 mL of distilled water to the mixing vessel as close as possible to the final volume.
  2. Add dry powder medium to room temperature (15 °C to 30 °C) water and stir gently. Do not heat the water.
  3. Rinse the inside of the package to remove any residual powder
  4. Add 3.024 g sodium bicarbonate/L medium.
  5. Adjust the final volume using distilled water, stirring until dissolved.
  6. Do not adjust the pH. Keep the vessel tightly closed until medium filtration is complete.
  7. Immediately use a positive pressure filtration system to process the medium into a sterile container through a 0.2 μm filtration membrane.
